Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007194.4(CHEK2):c.70A>C (p.Ser24Arg), citing Ambry Variant Classification Scheme 2023: The p.S24R variant (also known as c.70A>C), located in coding exon 1 of the CHEK2 gene, results from an A to C substitution at nucleotide position 70. The serine at codon 24 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
