Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000264.5(PTCH1):c.192G>C (p.Gln64His), citing Ambry Variant Classification Scheme 2023: The p.Q64H variant (also known as c.192G>C), located in coding exon 1 of the PTCH1 gene, results from a G to C substitution at nucleotide position 192. The glutamine at codon 64 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
