NM_000051.4(ATM):c.7921C>G (p.Gln2641Glu) was classified as Uncertain significance for Ataxia-telangiectasia syndrome by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals affected with ATM-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces glutamine with glutamic acid at codon 2641 of the ATM protein (p.Gln2641Glu). The glutamine residue is moderately conserved and there is a small physicochemical difference between glutamine and glutamic acid.
