Submitter rationale: The RB1 c.1922C>A (p.P641Q) variant has not been reported in the literature to our knowledge. It was observed in 1/8716 chromosomes of the African/African American subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). The variant has not been reported in ClinVar. Functional studies have not been performed, and in silico predictions of the variant's effect on protein function are inconclusive.
