Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003737.4(DCHS1):c.1583G>C (p.Gly528Ala), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine with alanine at codon 528 of the DCHS1 protein (p.Gly528Ala). The glycine residue is moderately conserved and there is a small physicochemical difference between glycine and alanine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with DCHS1-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt DCHS1 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
