Uncertain significance for Bardet-Biedl syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_015910.7(WDPCP):c.1055T>A (p.Leu352Gln), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals affected with WDPCP-related conditions. This variant is present in population databases (rs373387599, gnomAD 0.05%). This sequence change replaces leucine, which is neutral and non-polar, with glutamine, which is neutral and polar, at codon 352 of the WDPCP protein (p.Leu352Gln).
