Uncertain significance for Multiple endocrine neoplasia, type 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001370259.2(MEN1):c.1330C>G (p.Leu444Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine with valine at codon 444 of the MEN1 protein (p.Leu444Val). The leucine residue is moderately conserved and there is a small physicochemical difference between leucine and valine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt MEN1 protein function. This variant has not been reported in the literature in individuals with MEN1-related conditions. This variant is not present in population databases (ExAC no frequency).
