Uncertain significance for Leukodystrophy — the classification assigned by Broad Center for Mendelian Genomics, Broad Institute of MIT and Harvard to NM_007055.4(POLR3A):c.2563C>T (p.Arg855Trp), citing ACMG Guidelines, 2015: The p.Arg855Trp variant in POLR3A has been reported in 1 individual, in the compound heterozygous state, with POLR3A-related disorders (PMID: 34234304) and has been identified in 0.002% (1/34582) of Latino chromosomes by the Genome Aggregation Database (gnomAD, http://gnomad.broadinstitute.org; dbSNP ID: rs775160820). Although this variant has been seen in the general population in a heterozygous state, its frequency is low enough to be consistent with a recessive carrier frequency. This variant has also been reported in ClinVar (Variation ID: 1442721) and has been interpreted as a variant of uncertain significance by Invitae. Computational prediction tools and conservation analyses suggest that this variant may impact the protein, though this information is not predictive enough to determine pathogenicity.
