Uncertain significance for Symmetrical dyschromatosis of extremities; Aicardi-Goutieres syndrome 6 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001111.5(ADAR):c.2453C>G (p.Thr818Ser), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals affected with ADAR-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces threonine with serine at codon 818 of the ADAR protein (p.Thr818Ser). The threonine residue is weakly conserved and there is a small physicochemical difference between threonine and serine.
