Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_138393.4(REEP6):c.255G>C (p.Trp85Cys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tryptophan with cysteine at codon 85 of the REEP6 protein (p.Trp85Cys). The tryptophan residue is highly conserved and there is a large physicochemical difference between tryptophan and cysteine. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with REEP6-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
