NM_005670.4(EPM2A):c.937G>A (p.Ala313Thr) was classified as Uncertain significance for Progressive myoclonic epilepsy by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces alanine with threonine at codon 313 of the EPM2A protein (p.Ala313Thr). The alanine residue is highly conserved and there is a small physicochemical difference between alanine and threonine. This variant is present in population databases (rs189479757, ExAC 0.009%). This variant has not been reported in the literature in individuals affected with EPM2A-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive.
