Pathogenic for Isolated microphthalmia 5 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_031433.4(MFRP):c.1408C>T (p.Gln470Ter), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This sequence change creates a premature translational stop signal (p.Gln470*) in the MFRP gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in MFRP are known to be pathogenic (PMID: 12140190, 15976030, 20361016). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with MFRP-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site.
