Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000548.5(TSC2):c.3403C>T (p.His1135Tyr), citing Ambry Variant Classification Scheme 2023: The p.H1135Y variant (also known as c.3403C>T), located in coding exon 29 of the TSC2 gene, results from a C to T substitution at nucleotide position 3403. The histidine at codon 1135 is replaced by tyrosine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
