Pathogenic for Neuronopathy, distal hereditary motor, autosomal recessive 5 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006736.6(DNAJB2):c.119_120del (p.Glu40fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with DNAJB2-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Glu40Valfs*3) in the DNAJB2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in DNAJB2 are known to be pathogenic (PMID: 22522442, 25274842).
