Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001039958.2(MESP2):c.47G>A (p.Trp16Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Trp16*) in the MESP2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in MESP2 are known to be pathogenic (PMID: 9242490, 18485326). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with MESP2-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. For these reasons, this variant has been classified as Pathogenic.
