Uncertain significance for Congenital myotonia, autosomal dominant form; Congenital myotonia, autosomal recessive form — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000083.3(CLCN1):c.1006A>G (p.Asn336Asp), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the CLCN1 gene (transcript NM_000083.3) at coding-DNA position 1006, where A is replaced by G; at the protein level this means replaces asparagine at residue 336 with aspartic acid — a missense variant. Submitter rationale: This sequence change replaces asparagine with aspartic acid at codon 336 of the CLCN1 protein (p.Asn336Asp). The asparagine residue is highly conserved and there is a small physicochemical difference between asparagine and aspartic acid. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with CLCN1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C15"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
