This variant lies in the TCAP gene (transcript NM_003673.4) at coding-DNA position 387, where C is replaced by G; at the protein level this means replaces aspartic acid at residue 129 with glutamic acid — a missense variant. Submitter rationale: This sequence change replaces aspartic acid, which is acidic and polar, with glutamic acid, which is acidic and polar, at codon 129 of the TCAP protein (p.Asp129Glu). This variant is present in population databases (rs768942598, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with TCAP-related conditions. ClinVar contains an entry for this variant (Variation ID: 1441277).
