This variant lies in the RTEL1 gene (transcript NM_001283009.2) at coding-DNA position 436, where A is replaced by G; at the protein level this means replaces isoleucine at residue 146 with valine — a missense variant. Submitter rationale: This sequence change replaces isoleucine with valine at codon 146 of the RTEL1 protein (p.Ile146Val). The isoleucine residue is highly conserved and there is a small physicochemical difference between isoleucine and valine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with RTEL1-related conditions.
