Uncertain significance for Autosomal recessive limb-girdle muscular dystrophy type 2Q; Epidermolysis bullosa simplex, Ogna type; Epidermolysis bullosa simplex with nail dystrophy; Epidermolysis bullosa simplex 5C, with pyloric atresia; Epidermolysis bullosa simplex 5B, with muscular dystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_201384.3(PLEC):c.13631_13632del (p.Ser4544fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the PLEC gene (transcript NM_201384.3) at coding-DNA position 13631 through coding-DNA position 13632, deleting 2 bases; at the protein level this means shifts the reading frame starting at serine residue 4544, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change results in a frameshift in the PLEC gene (p.Ser4571Cysfs*40). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 4 amino acid(s) of the PLEC protein and extend the protein by 35 additional amino acid residues. This variant is present in population databases (no rsID available, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with PLEC-related conditions. ClinVar contains an entry for this variant (Variation ID: 1440833). Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
