NM_002528.7(NTHL1):c.242C>A (p.Pro81His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P89H variant (also known as c.266C>A), located in coding exon 2 of the NTHL1 gene, results from a C to A substitution at nucleotide position 266. The proline at codon 89 is replaced by histidine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear.
