Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001005373.4(LRSAM1):c.766T>C (p.Tyr256His), citing Ambry Variant Classification Scheme 2023: The c.766T>C (p.Y256H) alteration is located in exon 11 (coding exon 10) of the LRSAM1 gene. This alteration results from a T to C substitution at nucleotide position 766, causing the tyrosine (Y) at amino acid position 256 to be replaced by a histidine (H). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is highly conserved in available vertebrate species. The in silico prediction for this alteration is inconclusive. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
