NM_000722.4(CACNA2D1):c.3211T>C (p.Tyr1071His) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y1071H variant (also known as c.3211T>C), located in coding exon 39 of the CACNA2D1 gene, results from a T to C substitution at nucleotide position 3211. The tyrosine at codon 1071 is replaced by histidine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
