Uncertain significance for Bethlem myopathy 1A — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001849.4(COL6A2):c.2780C>G (p.Ala927Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces alanine with glycine at codon 927 of the COL6A2 protein (p.Ala927Gly). The alanine residue is weakly conserved and there is a small physicochemical difference between alanine and glycine. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt COL6A2 protein function. This variant is present in population databases (rs11554668, ExAC 0.04%). This variant has not been reported in the literature in individuals affected with COL6A2-related conditions.
