Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_033109.5(PNPT1):c.67C>A (p.Leu23Met), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine with methionine at codon 23 of the PNPT1 protein (p.Leu23Met). The leucine residue is weakly conserved and there is a small physicochemical difference between leucine and methionine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with PNPT1-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt PNPT1 protein function.
