This variant lies in the GNE gene (transcript NM_005476.7) at coding-DNA position 1162, where T is replaced by C; at the protein level this means replaces cysteine at residue 388 with arginine — a missense variant. Submitter rationale: This sequence change replaces cysteine with arginine at codon 419 of the GNE protein (p.Cys419Arg). The cysteine residue is highly conserved and there is a large physicochemical difference between cysteine and arginine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with GNE-related conditions.
