Uncertain significance for Metachromatic leukodystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000487.6(ARSA):c.1450G>C (p.Asp484His), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces aspartic acid, which is acidic and polar, with histidine, which is basic and polar, at codon 484 of the ARSA protein (p.Asp484His). This variant is present in population databases (rs749061034, gnomAD 0.005%). This variant has not been reported in the literature in individuals affected with ARSA-related conditions. ClinVar contains an entry for this variant (Variation ID: 1438541). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt ARSA protein function with a positive predictive value of 95%. Experimental studies have shown that this missense change affects ARSA function (PMID: 37480112).
