Pathogenic for Glycogen storage disease type III — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000642.3(AGL):c.2520_2524dup (p.Pro842fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Pro842Argfs*28) in the AGL gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in AGL are known to be pathogenic (PMID: 19299494). For these reasons, this variant has been classified as Pathogenic. This variant is also known as c.2524dupGTCTC (p.Leu867X). This premature translational stop signal has been observed in individual(s) with glycogen storage disease III (PMID: 19951495, 26984562). In at least one individual the data is consistent with the variant being in trans (on the opposite chromosome) from a pathogenic variant. This variant is present in population databases (rs759727721, ExAC 0.01%).
