This variant lies in the LRP4 gene (transcript NM_002334.4) at coding-DNA position 5140, where G is replaced by A; at the protein level this means replaces valine at residue 1714 with isoleucine — a missense variant. Submitter rationale: This variant has not been reported in the literature in individuals with LRP4-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0".
