Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This variant has not been reported in the literature in individuals affected with NPHP3-related conditions. This variant is present in population databases (rs762836818, ExAC 0.004%). This sequence change replaces glutamic acid with glutamine at codon 1013 of the NPHP3 protein (p.Glu1013Gln). The glutamic acid residue is highly conserved and there is a small physicochemical difference between glutamic acid and glutamine.
