Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004727.3(SLC24A1):c.2980G>A (p.Ala994Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces alanine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 994 of the SLC24A1 protein (p.Ala994Thr). This variant is present in population databases (rs200052767, gnomAD 0.008%). This missense change has been observed in individual(s) with clinical features of SLC24A1-related conditions (PMID: 12037007). ClinVar contains an entry for this variant (Variation ID: 1437252). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies have shown that this missense change does not substantially affect SLC24A1 function (PMID: 12037007). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
