Uncertain significance for LAMB2-related infantile-onset nephrotic syndrome; Pierson syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002292.4(LAMB2):c.5210A>C (p.Glu1737Ala), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This sequence change replaces glutamic acid with alanine at codon 1737 of the LAMB2 protein (p.Glu1737Ala). The glutamic acid residue is highly conserved and there is a moderate physicochemical difference between glutamic acid and alanine. This variant has not been reported in the literature in individuals affected with LAMB2-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive.
