Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000372.5(TYR):c.760G>A (p.Gly254Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine with serine at codon 254 of the TYR protein (p.Gly254Ser). The glycine residue is moderately conserved and there is a small physicochemical difference between glycine and serine. This variant is present in population databases (rs199500520, ExAC 0.009%). This variant has not been reported in the literature in individuals affected with TYR-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt TYR protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
