Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_024301.5(FKRP):c.101C>T (p.Ser34Phe), citing Ambry Variant Classification Scheme 2023: The p.S34F variant (also known as c.101C>T), located in coding exon 1 of the FKRP gene, results from a C to T substitution at nucleotide position 101. The serine at codon 34 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
