Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001110556.2(FLNA):c.7322G>A (p.Gly2441Asp), citing Ambry Variant Classification Scheme 2023: The c.7298G>A (p.G2433D) alteration is located in exon 44 (coding exon 43) of the FLNA gene. This alteration results from a G to A substitution at nucleotide position 7298, causing the glycine (G) at amino acid position 2433 to be replaced by an aspartic acid (D). Based on data from gnomAD, the A allele has an overall frequency of 0.005% (1/21897) total alleles studied. The highest observed frequency was 0.017% (1/5926) of African alleles. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
