Uncertain significance for Combined immunodeficiency due to DOCK8 deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_203447.4(DOCK8):c.590G>A (p.Cys197Tyr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces cysteine with tyrosine at codon 197 of the DOCK8 protein (p.Cys197Tyr). The cysteine residue is moderately conserved and there is a large physicochemical difference between cysteine and tyrosine. This variant is present in population databases (rs777075220, ExAC 0.002%). This variant has not been reported in the literature in individuals affected with DOCK8-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The tyrosine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function.
