Uncertain significance for Kufor-Rakeb syndrome; Autosomal recessive spastic paraplegia type 78 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_022089.4(ATP13A2):c.2326G>A (p.Val776Ile), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine with isoleucine at codon 776 of the ATP13A2 protein (p.Val776Ile). The valine residue is weakly conserved and there is a small physicochemical difference between valine and isoleucine. This variant is present in population databases (rs56170027, ExAC 0.01%). This variant has been observed in individual(s) with parkinsonism (PMID: 19085912). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt ATP13A2 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
