Uncertain significance for Joubert syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001173990.3(TMEM216):c.224T>G (p.Phe75Cys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces phenylalanine with cysteine at codon 75 of the TMEM216 protein (p.Phe75Cys). The phenylalanine residue is highly conserved and there is a large physicochemical difference between phenylalanine and cysteine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with TMEM216-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive.
