Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000843.4(GRM6):c.1357del (p.Ser453fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with GRM6-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Ser453Alafs*6) in the GRM6 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in GRM6 are known to be pathogenic (PMID: 15781871, 16622103, 22008250).
