Submitter rationale: The c.79_81dupAGC variant (also known as p.S27dup), located in coding exon 1 of the CBS gene, results from an in-frame duplication of AGC at nucleotide positions 79 to 81. This results in the duplication of an extra serine residue between codons 27 and 28. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
