This missense change has been observed in individual(s) with clinical features of neurofibromatosis type 1 (PMID: 16405917; Invitae). In at least one individual the variant was observed to be de novo. This sequence change replaces leucine with proline at codon 2082 of the NF1 protein (p.Leu2082Pro). The leucine residue is moderately conserved and there is a moderate physicochemical difference between leucine and proline.
