Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_005050.4(ABCD4):c.362G>A (p.Arg121His), citing LabCorp Variant Classification Summary - May 2015: Variant summary: ABCD4 c.362G>A (p.Arg121His) results in a non-conservative amino acid change located in the ABC transporter type 1, transmembrane domain (IPR011527) of the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 2.4e-05 in 250944 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.362G>A has been reported in the literature in an individual affected with Methylmalonic Acidemia With Homocystinuria without strong evidence of causality (Nair_2018). This report does not provide unequivocal conclusions about association of the variant with Methylmalonic Acidemia With Homocystinuria. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 30293248). Three submitters have cited clinical-significance assessments for this variant to ClinVar after 2014. All submitters classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
