Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001303256.3(MORC2):c.2963G>A (p.Arg988His), citing Ambry Variant Classification Scheme 2023: The p.R988H variant (also known as c.2963G>A), located in coding exon 25 of the MORC2 gene, results from a G to A substitution at nucleotide position 2963. The arginine at codon 988 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
