This variant lies in the PARN gene (transcript NM_002582.4) at coding-DNA position 1511, where A is replaced by G; at the protein level this means replaces tyrosine at residue 504 with cysteine — a missense variant. Submitter rationale: This sequence change replaces tyrosine with cysteine at codon 504 of the PARN protein (p.Tyr504Cys). The tyrosine residue is highly conserved and there is a large physicochemical difference between tyrosine and cysteine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with PARN-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
