This variant lies in the CARD11 gene (transcript NM_032415.7) at coding-DNA position 1084, where A is replaced by G; at the protein level this means replaces lysine at residue 362 with glutamic acid — a missense variant. Submitter rationale: This sequence change replaces lysine, which is basic and polar, with glutamic acid, which is acidic and polar, at codon 362 of the CARD11 protein (p.Lys362Glu). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with CARD11-related conditions. ClinVar contains an entry for this variant (Variation ID: 1431040).
