Pathogenic for Glycogen storage disease, type II — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000017.10:g.(?_78082486)_(78087175_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 8-15 of the GAA gene. This deletion is out-of-frame, and is expected to create a premature termination codon and result in an absent or disrupted protein product. Loss-of-function variants in GAA are known to be pathogenic (PMID: 18425781, 22252923). A similar copy number variant has been observed in individual(s) with glycogen storage disease (PMID: 11854868, 29122469). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is also known as IVS7-IVS15del. For these reasons, this variant has been classified as Pathogenic.
