Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001042545.2(LTBP4):c.4436G>A (p.Gly1479Asp), citing Invitae Variant Classification Sherloc (09022015): This variant is present in population databases (rs1051440, ExAC 0.01%). This sequence change replaces glycine with aspartic acid at codon 1509 of the LTBP4 protein (p.Gly1509Asp). The glycine residue is highly conserved and there is a moderate physicochemical difference between glycine and aspartic acid. This variant has not been reported in the literature in individuals affected with LTBP4-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown.
