NM_001085049.3(MRAS):c.378C>A (p.Asn126Lys) was classified as Uncertain significance for Idiopathic dilated cardiomyopathy; Noonan syndrome 11 by Clinical Genomics Laboratory, Stanford Medicine, citing ACMG Guidelines, 2015: The p.Asn126Lys variant in the MRAS gene has not been previously reported in association with disease. This variant was absent from large population databases, including the Genome Aggregation Database (http://gnomad.broadinstitute.org/). The asparagine at position 126 is evolutionarily conserved. Computational tools predict that the p.Asn126Lys variant is deleterious; however, the accuracy of in silico algorithms is limited. These data were assessed using the ACMG/AMP variant interpretation guidelines. In summary, the significance of the p.Asn126Lys variant is uncertain.
