Pathogenic for Carney-Stratakis syndrome; Paragangliomas with sensorineural hearing loss; Pheochromocytoma; Cowden syndrome 3 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003002.4(SDHD):c.288del (p.Ala97fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Ala97Glnfs*38) in the SDHD gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 63 amino acid(s) of the SDHD protein. For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the SDHD protein in which other variant(s) (p.Leu139Pro) have been determined to be pathogenic (PMID: 11391798, 21348866). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant has not been reported in the literature in individuals affected with SDHD-related conditions. This variant is not present in population databases (ExAC no frequency).
