Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_016030.6(TRAPPC12):c.250G>T (p.Asp84Tyr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces aspartic acid with tyrosine at codon 84 of the TRAPPC12 protein (p.Asp84Tyr). The aspartic acid residue is weakly conserved and there is a large physicochemical difference between aspartic acid and tyrosine. This variant is present in population databases (rs529746013, gnomAD 0.07%). This variant has not been reported in the literature in individuals affected with TRAPPC12-related conditions. ClinVar contains an entry for this variant (Variation ID: 1430336).
