NM_001127222.2(CACNA1A):c.4634G>A (p.Arg1545Gln) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R1546Q variant (also known as c.4637G>A), located in coding exon 29 of the CACNA1A gene, results from a G to A substitution at nucleotide position 4637. The arginine at codon 1546 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
